Jamie Ward (hamstring) has shaken off the injury that ruled him out of international action.
James O'Connor (knee) misses out, while Mason Bennett is away with England Under-17s in Portugal.
Defender Michael Hoganson is a late addition to the bench, as a result of O'Brien's withdrawal.
Derby have lost only once in 10 visits to Hillsborough - and are unbeaten in four.
However, the Owls are in good form, having only suffered one defeat in nine league outings.
Wednesday manager Dave Jones has named an unchanged side, with new loan striker Connor Wickham on the bench, along with former Rams keeper Stephen Bywater.
